Alkaline Phosphatase, Tissue-Nonspecific Isozyme (TNAP) is a ubiquitously expressed enzyme encoded by the ALPL gene. As the predominant alkaline phosphatase in humans, it is crucial for bone and tooth mineralization. TNAP hydrolyzes inorganic pyrophosphate, a potent mineralization inhibitor, and provides inorganic phosphate to promote hydroxyapatite crystal growth. Beyond its skeletal role, TNAP is also vital in neuronal development, vitamin B6 metabolism, and modulating intestinal inflammation. Mutations in ALPL cause hypophosphatasia, a metabolic bone disorder characterized by defective mineralization. Thus, TNAP serves as a central regulator of phosphate metabolism across multiple physiological systems.
