The Biotin-KRAS[G12A] activity was detected using HTRF technology. The reaction was performed by incubating the Biotin-KRAS[G12A] protein, GTP and SOS1 at 25℃ for 60 min, adding cRAF at 25℃ for 15 min and adding the beads and keeping the mixture at 25℃ for 60 min, then reading ratio 665/620 with BMG.
