背景介绍

  • O-Glycoproteinase is expressed in E. coli and is a highly specific glycoproteinase that can specifically recognize serine or threonine residues of mucin-type O-linked sugars (whether sialylated or not) and cleave them at their N-termini. It can produce O-glycopeptides suitable for mass spectrometry analysis. The enzyme contains a 6X His-Tag, which can be easily removed after the reaction.

操作步骤

  • 仅供参考：

    • 将10µg糖蛋白与20 mM Tris-HCl pH 8.0, 40 mM DTT, 0.1% SDS溶液混合，并使得总体积为50µl。
    • 95°C加热10分钟。
    • 冷却至室温。
    • 加入3µl 500mM 碘乙酸溶液。
    • 室温放置30分钟。
    • 将溶液转移至超滤离心管中。
    • 12000 x g 离心4分钟，弃去流穿。
    • 加入400µl 20mM Tris-HCl，pH 8.0溶液。
    • 12000 x g 离心4分钟，弃去流穿。加入400µl 20mM Tris-HCl，pH 8.0溶液，重复一次。
    • 加入20µl 20mM Tris-HCl，pH 8.0溶液。
    • 加入1µl O糖蛋白酶，轻柔混匀。
    • 37˚C孵育2-18小时。
    • 12000 x g 离心4分钟，收集上清。

  • One unit is the amount of enzyme required to cleave more than 90% of a 2 µM O-glycopeptide in 20 mM Tris-HCl, pH 8.0 in a 20 μL reaction at 37°C for 2 hours.
