Interleukin-27 (IL-27) is a heterodimeric cytokine belonging to the IL-12 family. It is composed of two subunits, Epstein-Barr virus-induced gene 3 (EBI3) and p28, and signals through a receptor complex formed by WSX-1 (also known as TCCR or IL-27Rα) and gp130.
IL-27 is produced primarily by antigen-presenting cells (APCs), such as monocytes, dendritic cells, and macrophages, upon activation of pattern recognition receptors. The cynomolgus monkey (Macaca fascicularis) IL-27 protein shares a high degree of amino acid identity with its human ortholog (e.g., 92% for the p28 subunit), enabling the translation of findings from preclinical models.
The biological effects of IL-27 are pleiotropic and context-dependent, integrating both pro-inflammatory and anti-inflammatory functions. It plays a key role in the early induction of Th1 differentiation and also exerts immunosuppressive effects. The specific outcome of IL-27 signaling—whether it promotes or resolves inflammation—is determined by the cellular context, disease state, and infection phase.
