Glia Maturation Factor beta (GMFB) is a 17 kDa intracellular protein primarily expressed in the brain's glial cells, though it is also present in other tissues. As a member of the actin-depolymerizing factor family, it functions as a crucial intracellular regulator of actin cytoskeleton dynamics and cellular signaling pathways (notably enhancing p38 MAPK and inhibiting ERK). Its dysregulation is a key amplifier of inflammatory responses and oxidative stress, linking it to the pathology of numerous diseases.
GMFB is a major driver in diseases like Alzheimer's, Parkinson's, and Multiple Sclerosis, where it promotes glial activation and cytokine release. It is considered a promising therapeutic target; studies show GMFB deficiency protects against neuroinflammation.
