APRIL (A Proliferation-Inducing Ligand), also known as TNFSF13, is a member of the tumor necrosis factor (TNF) superfamily encoded by the TNFSF13 gene located on human chromosome 17p13. This 250-amino acid type II transmembrane protein functions as a homotrimeric cytokine and plays pivotal roles in immune regulation, B cell biology, and various pathological conditions including cancer, autoimmune diseases, and cardiovascular disorders.
APRIL is extensively studied in systemic lupus erythematosus (SLE), Sjögren's syndrome, IgA nephropathy, and rheumatoid arthritis. Elevated APRIL levels correlate with disease activity, making it a therapeutic target. Atacicept (TACI-Ig fusion protein) neutralizes both APRIL and BAFF, showing promise in clinical trials.
