背景介绍

  • HB-EGF is a member of the epidermal growth factor (EGF) family, encoded by the HBEGF gene on human chromosome 5q23. It is synthesized as a type I transmembrane precursor (pro-HB-EGF) of approximately 208 amino acids, which undergoes proteolytic cleavage by ADAM10 and ADAM17 metalloproteinases to release a soluble 87-amino acid mature form (sHB-EGF). Notably, the remaining C-terminal fragment can translocate to the nucleus and regulate transcription, representing a novel signaling mechanism distinct from canonical EGFR activation.
    HB-EGF uniquely contains a heparin-binding domain that mediates high-affinity interactions with heparan sulfate proteoglycans (HSPGs), enabling localization to cell-cell contact sites and modulating its bioavailability. This juxtacrine signaling property distinguishes HB-EGF from other EGF family members.
