Interleukin-17A (IL-17A), also referred to as cytotoxic T-lymphocyte-associated antigen 8 (CTLA8), is a key proinflammatory cytokine synthesized by activated T cells. It exerts its biological effects by regulating the activities of NF-kappaB and mitogen-activated protein kinases, which are critical in inflammatory signaling pathways. Additionally, IL-17A can induce the expression of IL6 and cyclooxygenase-2 (PTGS2/COX-2), while also promoting the production of nitric oxide (NO), all of which contribute to amplifying inflammatory responses. This cytokine exists in two forms: glycosylated and nonglycosylated, each with potential differences in biological activity. Notably, elevated levels of IL-17A have been closely linked to the pathogenesis of various chronic inflammatory disorders. These include rheumatoid arthritis, where it drives joint inflammation, psoriasis, contributing to skin lesion formation, and multiple sclerosis, involved in neuroinflammation. Understanding the role of IL-17A in these conditions has become a focus of research for developing targeted therapeutic strategies.
