Ofatumumab is a fully humanized monoclonal antibody that targets the CD20 molecule on the surface of B cells. Its core mechanism of action involves binding to the CD20 protein, thereby triggering multiple effector functions—including antibody-dependent cellular cytotoxicity, complement-dependent cytotoxicity, and apoptosis—to efficiently deplete CD20-positive B lymphocytes. Clinically, ofatumumab has a dual application: the intravenous formulation (Arzerra®) is indicated for the treatment of chronic lymphocytic leukemia, while the subcutaneous formulation (Kesimpta®) has been approved for the management of adult relapsing forms of multiple sclerosis, effectively reducing relapse rates and delaying disability progression. Notably, its convenient once‑monthly, at‑home self‑administration regimen offers significant advantages in facilitating long‑term disease management for patients with multiple sclerosis.
