背景介绍

  • CDCP1 (CUB Domain Containing Protein 1, also known as SIMA135 or TRASK) is a single-pass transmembrane glycoprotein featuring three extracellular CUB domains and proteolytic cleavage sites; its intracellular segment activates Src family kinases (SFKs) via Y734 phosphorylation, thereby regulating cell adhesion, motility, and anoikis resistance. This protein can bind to integrin β1 to activate FAK/PI3K/Akt signaling and functions as a hypoxia-inducible factor-2α (HIF-2α) target gene under hypoxic regulation; in breast cancer and renal cell carcinoma, CDCP1 drives metastasis through promoting fatty acid oxidation and hypoxic adaptation, respectively. Clinical studies demonstrate that CDCP1 is highly expressed in melanoma, pancreatic cancer, prostate cancer, and other solid tumors, correlating with poor patient prognosis and castration-resistant prostate cancer progression, establishing it as a potential prognostic biomarker and therapeutic target, with anti-CDCP1 antibodies (such as 4A06) currently being investigated for theranostic targeted therapy applications, offering novel precision intervention strategies for metastatic malignancies.
