背景介绍
  • This fivecolor panel is designed for detecting myeloidderived suppressor cell (MDSC) subsets in mouse tissue cells. CD11b was included as a general myeloid cell marker, and Ly6G/Ly6C (Gr1) was included to identify the total myeloid population . Ly6C and Ly6G were then used to distinguish monocytic MDSCs (MMDSC) and granulocytic MDSCs (GMDSC). Finally, CD115 (the MCSF receptor) was included to further characterize myeloid cells, as its expression can provide additional information on monocyterelated populations or MDSC differentiation stages. The panel is compatible with most twolaser flow cytometers. If desired, the panel can serve as a backbone panel, allowing for the addition of antibodies conjugated to various fluorophores to detect other markers of interest.

    • Mouse bone marrow were stained using the Mouse MDSCs Panel Kit consisting of anti-mouse CD11b PerCP-Cy5.5, anti-mouse Ly-6G/Ly-6C (Gr-1) Pacific Blue, anti-mouse Ly6C FITC, anti-mouse Ly6G APC and anti-mouse CD115 PE conjugated antibodies.
      To analyze the data:
      1. Create an FSC-A vs SSC-A dot plot and draw a gate around the leukocyte population.
      2. Create a Fixable Viability Dye 545 (S0B88802) vs SSC-A dot plot with the leukocyte gate applied. Draw a gate around the live cell population.
      3. Create a CD11b PerCP-Cy5.5-A vs Ly-6G/Ly-6C (Gr-1) Pacific Blue-A dot plot show the myeloid cells with the live cells gate applied.
      4. Create a Ly6G APC-A vs Ly6C FITC-A dot plot with the myeloid cells gate applied. Create gates to encompass Ly6G-Ly6Chi cells (M-MDSC), Ly6G+Ly6Gmid cells (G-MDSC)
      5. Create a CD115 PE-A vs SSC-A dot plot with the CD11b+ gate applied.
