Phospho-Stat3 (Tyr705) is a critical biomarker indicating the activation of the STAT3 protein, a latent cytoplasmic transcription factor involved in various cellular processes such as proliferation, survival, and immune response. When phosphorylated at tyrosine 705, STAT3 undergoes dimerization and translocates to the nucleus, where it binds to DNA and regulates gene expression. This phosphorylation is primarily mediated by the JAK family of receptor-associated tyrosine kinases, particularly JAK1, in response to cytokine signaling. Phospho-Stat3 (Tyr705) is also implicated in diseases like cancer, where its constitutive activation is observed in various tumor types.
