Disitamab Vedotin is a HER2-targeted antibody-drug conjugate (ADC). It consists of an antibody designed to specifically bind to the HER2 protein on tumor cells (with high affinity and to a different epitope than trastuzumab), a cleavable linker, and a potent cytotoxic agent called monomethyl auristatin E (MMAE). Its mechanism of action resembles a "biological missile": after binding to HER2, the entire molecule is internalized into the tumor cell. The linker is then cleaved intracellularly, releasing MMAE, which disrupts the cell's microtubule structure, ultimately leading to tumor cell death.
