PVRIG is a co-inhibitory receptor belonging to the poliovirus receptor (PVR) family. It is a 36kD single-pass transmembrane protein composed of one transmembrane domain, a long intracellular domain, and a single extracellular immunoglobulin variable-like (IgV) domain. PVRIG is predominantly expressed on natural killer (NK) cells and T cells, especially CD8+ T cells, and its expression is upregulated upon activation. In the tumor microenvironment, PVRIG is highly expressed on NK cells, CD4+ T cells, and CD8+ T cells. It binds to its ligand CD112 (NECTIN2 or PVRL2) with high affinity, inhibiting the activation of T cells and NK cells, thus contributing to immune evasion in cancer. This makes the PVRIG /CD112 axis a potential target for cancer immunotherapy.
