  • Rat alpha 2-Macroglobulin (A2M) is a large plasma protein that functions as a broad-spectrum protease inhibitor. It is primarily synthesized in the liver and circulates in the blood as a homotetrameric complex. Unlike smaller protease inhibitors, A2M inhibits proteases from all four catalytic classes by a unique entrapment mechanism. Upon cleavage of its bait region, the protein undergoes a major conformational change that physically encloses the target protease. This entrapped enzyme remains active toward small substrates but is blocked from accessing large protein substrates. Rat A2M also binds and facilitates the clearance of various growth factors, cytokines, and toxins through receptor-mediated endocytosis via the low-density lipoprotein receptor-related protein (LRP). Thus, it plays a critical role in modulating inflammatory responses, tissue remodeling, and host defense in rats.
