Human CCL26, also known as Eotaxin-3, is a small cytokine belonging to the CC chemokine family, primarily involved in allergic inflammation and eosinophil recruitment. Encoded by the CCL26 gene located on chromosome 7q11.23, this protein is secreted by endothelial cells, epithelial cells, and fibroblasts, particularly in response to stimulation by IL-4 and IL-13. Its primary function is to bind specifically to the chemokine receptor CCR3, which is highly expressed on eosinophils, basophils, and Th2 lymphocytes, thereby mediating their chemotaxis to sites of allergic inflammation. Unlike its homolog Eotaxin-1 (CCL11), CCL26 is selectively induced by Th2-associated cytokines and plays a prominent role in chronic allergic conditions such as atopic dermatitis, asthma, and eosinophilic esophagitis. Elevated expression of CCL26 has been observed in diseased tissues and correlates with eosinophil infiltration and disease severity. Due to its specific role in mediating type 2 inflammatory responses, CCL26 is considered both a diagnostic biomarker and a potential therapeutic target for allergic disorders.
