The Magnetic Bead-Based Immunoprecipitation (IP) Kit utilizes superparamagnetic Protein A/G conjugated magnetic beads for the specific isolation and purification of target antigens, antigen-antibody complexes, or protein complexes from biological samples such as cell lysates and tissue homogenates. Leveraging the high specific surface area and efficient magnetic separation properties of the beads, the kit enables rapid enrichment of target proteins without centrifugation, offering advantages including simple operation, low background, good reproducibility, and high yield. Two elution methods (acidic elution and denaturing elution) are provided to meet different experimental requirements: preserving protein activity or achieving complete elution. It is suitable for immunoprecipitation (IP), co-immunoprecipitation (Co-IP), Western blot analysis, mass spectrometry, and protein-protein interaction studies.

Immunoprecipitation (IP) is a technique based on the specific antigen-antibody binding reaction. The magnetic beads in this kit are covalently conjugated with protein A/G, which can specifically bind to the Fc region of immunoglobulins (IgG) from various species. The workflow involves the following key steps: 1) Add protein A/G- magnetic beads to capture antibody through the interaction between protein A/G and the antibody's Fc region; 2) Incubate the sample (containing the target antigen) with the beads-antibody complex to form beads-antigen-antibody complexes; 3) Wash the beads to remove non-specifically bound impurities; 4) Elute the target antigen (or antigen-antibody complex) from the beads using either acidic or denaturing elution buffer; 5) Collect the eluted sample for subsequent experimental analysis.
