The Agarose Bead-Based Immunoprecipitation (IP) Kit is designed for the specific isolation and purification of antigens, antigen-antibody complexes, or other target proteins from cell lysates, tissue homogenates, or other biological samples using agarose beads conjugated with protein A/G. This kit provides a comprehensive set of reagents and optimized protocols to ensure efficient immunoprecipitation with high specificity and low background. Two elution methods (acidic elution and denaturing elution) are included to meet different experimental requirements, such as preserving protein activity (acidic elution) or achieving complete elution for subsequent analysis (denaturing elution).

Immunoprecipitation (IP) is a technique based on the specific antigen-antibody binding reaction. The agarose beads in this kit are covalently conjugated with protein A/G, which can specifically bind to the Fc region of immunoglobulins (IgG) from various species. The workflow involves the following key steps: 1) Add protein A/G-agarose beads to capture antibody through the interaction between protein A/G and the antibody's Fc region; 2) Incubate the sample (containing the target antigen) with the beads-antibody complex to form beads-antigen-antibody complexes; 3) Wash the beads to remove non-specifically bound impurities; 4) Elute the target antigen (or antigen-antibody complex) from the beads using either acidic or denaturing elution buffer; 5) Collect the eluted sample for subsequent experimental analysis.
