背景介绍
  • CD32b/c (also known as FcγRIIb/c) is a low-affinity receptor for the Fc region of IgG antibodies, expressed primarily on B cells, mast cells, basophils, and some dendritic cells and macrophages. As an inhibitory Fc receptor, CD32b contains an immunoreceptor tyrosine-based inhibition motif (ITIM) in its cytoplasmic tail that delivers negative signals upon cross-linking with immune complexes, thereby suppressing antibody production, preventing excessive immune activation, and maintaining immune tolerance. CD32c, a highly homologous variant differing by only a few amino acids in the extracellular domain, lacks the ITIM and instead associates with the FcRγ chain to mediate activating signals, though its expression is more restricted and less characterized compared to CD32b. Together, these receptors play critical roles in regulating immune complex clearance, autoantibody production, and the balance between protective immunity and autoimmune pathology.

  • 流式分析

    • Flow cytometric analysis of human peripheral blood lymphocytes labelling Human CD32b/c antibody at 1/200 dilution (1μg) / (right panel) was compared with a Mouse IgG1, κ Isotype Control / (left panel). Goat Anti-Mouse IgG Alexa Fluor® 488 was used as the secondary antibody. Then cells were stained with CD19 - Brilliant Violet 421™ antibody separately. Total viable cells, as determined by Fixable Viability Dye 662 (S0D0016), were used for analysis. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.
