CD42a, also known as glycoprotein IX (GPIX), is an essential subunit of the platelet surface glycoprotein Ib-IX-V complex. This complex serves as the primary receptor for von Willebrand factor (vWF) and plays a central role in platelet adhesion, thrombus formation, and primary hemostasis. CD42a non-covalently associates with other subunits such as glycoprotein Ibα (GPIbα) and glycoprotein Ibβ (GPIbβ) to maintain the structural stability and proper membrane expression of the receptor complex. Upon vascular injury, the CD42a-containing complex rapidly binds exposed vWF, mediating firm platelet adhesion to the subendothelial matrix under high shear stress and initiating downstream signaling that leads to platelet activation and aggregation. Mutations in the CD42a gene (GP9) cause the rare autosomal recessive disorder Bernard-Soulier syndrome, characterized by giant platelets, thrombocytopenia, and a severe bleeding tendency. Furthermore, as a platelet-specific marker, CD42a holds significant value in flow cytometry, quality assessment of platelet transfusion, and research into the pathological mechanisms of thrombotic disorders.
流式分析
Flow cytometric analysis of human CD42a expression on fresh human peripheral blood cells. Fresh human peripheral blood cells were stained with Allophycocyanin Mouse Anti-Human CD41 Antibody and either FITC Mouse IgG1, κ Isotype Control (left panel) or SDT FITC Mouse Anti-Human CD42a Antibody (right panel) at 5 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.
