背景介绍
  • CD226, also known as DNAM-1, is a transmembrane glycoprotein in the immunoglobulin superfamily that functions as a key co-stimulatory receptor on T cells, NK cells, monocytes, and platelets, where it binds the nectin-family ligands CD155 and CD112 to trigger phosphorylation of its intracellular tyrosine and serine residues, thereby recruiting LFA-1 and activating Src, PI3K-AKT, VAV1-ERK, and PLCγ2 signaling cascades that enhance immune synapse formation, cytotoxic granule release, and production of IFN-γ and other cytokines, thus promoting antiviral and antitumor immunity; its expression correlates with improved outcomes in cancer immunotherapy, while its genetic polymorphisms or down-regulation are linked to autoimmune diseases and tumor immune evasion, making CD226 an emerging target for immunomodulatory therapies.

  • 流式分析

    • Flow cytometric analysis of Mouse CD226 expression on BALB/c mouse splenocytes. BALB/c mouse splenocytes were stained with Alexa Fluor® 488 Rat Anti-Mouse CD8α Antibody (S0B8041) and either Biotin Rat IgG2b, k Isotype Control (left panel) or SDT Biotin Rat Anti- Mouse CD226 Antibody (right panel) at 2 μl/test followed by Sav-iFluor 488. Total viable cells, as determined by Fixable Viability Dye 452 (S0D0021), were used for analysis. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.
