CD21, also known as complement receptor type 2 (CR2), is a ~145 kDa membrane glycoprotein expressed mainly on mature B cells and follicular dendritic cells that functions as the principal cellular receptor for complement fragments such as C3d and for the Epstein–Barr virus gp350/220 glycoprotein, thereby bridging innate and adaptive immunity by forming the CD19/CD21/CD81 coreceptor complex that lowers the B-cell receptor signaling threshold, promotes B-cell activation, proliferation, antibody production and antigen presentation, and also participates in retaining immune complexes in germinal centers, while its soluble ectodomain (sCD21) released by metalloprotease shedding circulates as an immunomodulatory decoy that can either enhance or suppress immune responses depending on context and is dysregulated in various infectious, neoplastic and autoimmune disorders.

Flow cytometric analysis of Human CD21 expression on Human PBMC (human peripheral blood mononuclear cells). Cells from the Human PBMC was stained with Brilliant Violet 421™ Mouse Anti-Human CD19 and either Alexa Fluor® 700 Mouse IgG1, κ Isotype Control (Left panel) and SDT Alexa Fluor® 700 Mouse Anti-Human CD21 Antibody (Right panel) at 5 μl/test. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.
