CHCHD2 (Coiled-Coil-Helix-Coiled-Coil-Helix Domain Containing Protein 2, also known as MNRR1 or PARK22) is a small nuclear-encoded protein primarily localized in the mitochondrial intermembrane space. It belongs to the protein family containing a conserved CX₉C motif, which folds into a characteristic CHCH domain via disulfide bond formation. This protein plays a central role in maintaining mitochondrial homeostasis: it acts both as a regulator of oxidative phosphorylation (particularly cytochrome c oxidase), helping to stabilize mitochondrial cristae structure, and as a transcriptional coactivator that translocates to the nucleus under cellular stress to regulate the expression of hypoxia-related genes. Additionally, by interacting with BCL-2 family proteins, CHCHD2 functions as a negative regulator of mitochondria-mediated apoptosis, exerting anti-apoptotic effects. On the pathological front, dominant point mutations in the CHCHD2 gene (e.g., T61I) have been confirmed as a causative factor for autosomal dominant Parkinson's disease: the mutations lead to protein misfolding and aggregation within mitochondria, subsequently triggering mitochondrial respiratory chain dysfunction, accumulation of reactive oxygen species, and promoting the abnormal aggregation of α-synuclein, ultimately resulting in the functional impairment of dopaminergic neurons. This gain-of-function toxic mechanism positions CHCHD2 as a key molecular node linking mitochondrial dysfunction to the pathological processes of neurodegenerative diseases, making it a potential target for precision therapy.
