Ribosomal S6 Kinases 1, 2, and 3 (RSK1, RSK2, and RSK3) are serine/threonine kinases belonging to the p90 ribosomal S6 kinase (p90RSK) family, which function as critical downstream effectors of the Ras-MAPK/ERK signaling pathway to regulate diverse cellular processes including proliferation, survival, differentiation, and metabolism. These isoforms share a unique bimodal structure comprising an N-terminal kinase domain and a C-terminal kinase domain linked by a regulatory loop, where activation typically involves phosphorylation by ERK1/2 at the C-terminal domain followed by autophosphorylation and subsequent activation of the N-terminal domain to phosphorylate various substrates such as CREB, Bad, and GSK3β. While they exhibit high sequence homology and overlapping functions, RSK1, RSK2, and RSK3 display distinct tissue distribution patterns and specific physiological roles; for instance, RSK2 mutations are directly linked to Coffin-Lowry syndrome, a rare X-linked genetic disorder characterized by severe psychomotor retardation and skeletal abnormalities, highlighting the non-redundant importance of specific RSK isoforms in neural development and cognitive function, whereas RSK1 and RSK3 are often implicated in cancer progression and cardiac hypertrophy, making them significant targets for therapeutic intervention in oncology and cardiovascular diseases.
