Fibroblast Growth Factor 18 (FGF18) is a secreted signaling protein belonging to the fibroblast growth factor family, playing pivotal roles in skeletal development, tissue repair, and cellular proliferation. It primarily exerts its biological effects by binding to specific FGF receptors (particularly FGFR3 and FGFR4) in the presence of heparan sulfate proteoglycans, thereby activating downstream pathways such as MAPK and PI3K/Akt. FGF18 is critically involved in endochondral ossification, where it regulates chondrocyte proliferation and differentiation during bone formation, making it essential for proper long bone growth. Beyond skeletal biology, it contributes to liver regeneration, angiogenesis, and wound healing, and has emerged as a promising therapeutic candidate for treating osteoarthritis and promoting cartilage repair due to its anabolic effects on joint tissues.
