Mouse C3 protein, the murine ortholog of the human complement component 3, is a pivotal glycoprotein synthesized primarily by hepatocytes and macrophages that serves as the central hub of the complement system, playing an indispensable role in both innate and adaptive immunity. Structurally composed of an alpha and a beta chain linked by disulfide bonds, C3 circulates in the blood as an inactive zymogen until it is cleaved by C3 convertases into two biologically active fragments: C3a, a potent anaphylatoxin that recruits inflammatory cells and induces mast cell degranulation, and C3b, which acts as a major opsonin to tag pathogens for phagocytosis and facilitates the assembly of the membrane attack complex (MAC) for direct cell lysis. Beyond its canonical immune functions, mouse C3 is critically involved in synaptic pruning during central nervous system development, where microglia utilize C3-tagged synapses to eliminate redundant neural connections, and it has been extensively studied in various murine disease models—including autoimmune disorders, neurodegenerative diseases like Alzheimer’s, and renal pathologies—to elucidate the mechanistic underpinnings of complement-mediated tissue damage and regeneration, making it a vital subject for understanding immunological homeostasis and therapeutic targeting in preclinical research.
