Lymphatic Vessel Endothelial Hyaluronan Receptor 1 (LYVE-1) is a type I integral membrane glycoprotein belonging to the Link protein superfamily, primarily characterized by its high-affinity binding to hyaluronan (HA), a major component of the extracellular matrix. Predominantly expressed on the surface of lymphatic endothelial cells in both developing and adult tissues, LYVE-1 serves as a definitive marker for identifying lymphatic vessels, distinguishing them from blood vessels, although it is also transiently found on liver sinusoidal endothelial cells and certain macrophage populations during embryogenesis or inflammation. Structurally, it shares homology with CD44, another HA receptor, yet possesses unique functional properties that facilitate the uptake and transport of HA from peripheral tissues into the lymphatic system, thereby playing a critical role in maintaining tissue fluid homeostasis, regulating immune cell trafficking, and modulating inflammatory responses. Beyond its physiological functions in lymphangiogenesis and HA clearance, LYVE-1 has significant clinical relevance as its expression is often upregulated in tumor-associated lymphatics, making it a valuable target for studying cancer metastasis and a potential diagnostic marker for lymphatic malformations and various carcinomas.

IHC shows positive staining in paraffin-embedded human colon. Anti-LYVE1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
