Spry2 (Sprouty homolog 2) is a critical intracellular negative regulator of signal transduction that functions primarily as a feedback inhibitor downstream of receptor tyrosine kinase (RTK) pathways, such as FGF, EGF, VEGF, and MET; by utilizing its N-terminal cysteine-rich domain for membrane localization and its C-terminal domain to interact with signaling molecules like Grb2, Sos, or Raf, Spry2 effectively blocks the activation of the Ras-MAPK/ERK cascade. This protein plays an essential spatiotemporal regulatory role during embryonic development, particularly in limb formation, branching morphogenesis (e.g., in the lung, kidney, and salivary glands), and angiogenesis, ensuring that growth factor signaling intensity and duration remain within physiological limits, while its aberrant expression or functional loss is closely associated with the onset and progression of various cancers, tumor angiogenesis, and drug resistance.
