  • BCAR1 (Breast Cancer Anti-Estrogen Resistance 1), also known as p130Cas (Crk-associated substrate), is a multifunctional adaptor/scaffold protein that plays a pivotal role in integrating signals from integrins, growth factor receptors, and other cellular stimuli to regulate cell adhesion, migration, proliferation, and survival. Structurally, BCAR1 contains an N-terminal SH3 domain, a proline-rich region, a large substrate domain with multiple YxxP motifs that serve as phosphorylation sites for Src family kinases, and a C-terminal focal adhesion targeting (FAT) domain that mediates its localization to focal adhesions through interactions with paxillin and other proteins. Upon tyrosine phosphorylation, BCAR1 acts as a docking platform for various signaling molecules including Crk, Nck, and PI3K, thereby activating downstream pathways such as Ras-MAPK and PI3K-AKT that promote cytoskeletal reorganization and oncogenic transformation. Aberrant overexpression or hyperactivation of BCAR1 has been strongly implicated in tumor progression, metastasis, and resistance to endocrine therapies in breast cancer and other malignancies, making it a potential prognostic biomarker and therapeutic target.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

    • ICC shows positive staining in NIH/3T3 cells. Anti-BCAR1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C.
