PDK1 (officially PDPK1), the 3-phosphoinositide-dependent protein kinase-1, is a 556-aa serine/threonine kinase whose N-terminal catalytic domain followed by a C-terminal PH domain that binds PtdIns(3,4,5)P3 and PtdIns(3,4)P2 generated by PI3K; once recruited to the plasma membrane it uses a docking-site motif (termed PIF-pocket) to phosphorylate the T-loop of AGC kinases such as AKT, S6K, SGK and PKC isoforms, thereby serving as the master upstream activator of the PI3K–AKT–mTOR signaling axis that controls cell growth, proliferation, survival and metabolism, while its dysregulation through PTEN loss, PI3K mutation or PDK1 overexpression is a frequent driver of cancer and metabolic disorders.

免疫细胞化学
ICC shows positive staining in HeLa cells. Anti- PDK1/PDPK1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
