背景介绍
  • VE-Cadherin (vascular endothelial cadherin, CDH5) is a 140-kDa transmembrane glycoprotein that forms the core of endothelial adherens junctions, where its calcium-dependent homophilic interactions between extracellular EC1-EC5 repeats create a semi-permeable barrier between blood and tissues; its cytoplasmic tail recruits β-catenin, plakoglobin and p120-catenin, linking the junction to the actin cytoskeleton and enabling transmission of shear-stress signals that regulate angiogenesis, vascular remodeling and leukocyte extravasation, while phosphorylation of Y685 and Y658 by Src, VEGFR2 or Tie-2 and subsequent internalization of the complex transiently increases permeability during inflammation, tumor metastasis and atherosclerosis, making VE-cadherin a key therapeutic target in vascular pathologies.

    • ICC shows positive staining in bEnd.3 cells (top panel) and negative staining in NIH/3T3 cells (below panel). Anti-VE-Cadherin antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
