RORγt is a DNA-binding transcription factor of the nuclear-receptor superfamily that serves as the master lineage-defining factor for Th17 cells, ILC3s, and other IL-17-producing lymphocytes, where it integrates cholesterol-derived oxysterol signals to drive the transcription of IL-17A/F, IL-22, and related pro-inflammatory genes, thereby orchestrating mucosal host defense against extracellular bacteria and fungi while also controlling circadian gene networks, lymphoid-organ development, and Treg/Th17 balance, making its activity—tunable by small-molecule agonists or inverse agonists—a pivotal rheostat in autoimmune, neoplastic, and infectious disease pathogenesis.
