RanBP2, a 358-kDa nucleoporin that forms the cytoplasmic filaments of the nuclear pore complex, functions as an E3 SUMO ligase and a scaffold for RanGAP1-SUMO1-Ubc9 complexes, thereby orchestrating nucleocytoplasmic transport, mitotic spindle assembly, and the recruitment of kinesin-1 to drive plus-end-directed motility of aggresomes and lipid droplets while also regulating NF-κB signaling, circadian rhythms, and neuronal responses through its extensive network of interactions at the pore and beyond.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

ICC shows positive staining in HeLa cells. Anti-RanBP2 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
