背景介绍

  • DCBLD2 (Discoidin, CUB and LCCL domain-containing protein 2) is a type-I transmembrane scaffolding receptor encoded on chromosome 3q12.1 that contains extracellular CUB, LCCL and coagulation factor V/VIII homology domains, seven conserved intracellular YxxP phosphorylation motifs, and a C-terminal PDZ-binding motif, enabling it to act as a multi-functional platform coupling receptor tyrosine kinases (e.g., EGFR) or non-receptor tyrosine kinases (e.g., Src, Abl) to downstream signaling cascades such as TRAF6-Akt, focal-adhesion/ITGB1-FAK and CRKL pathways, thereby regulating vascular endothelial and smooth-muscle-cell proliferation, angiogenesis, epithelial-to-mesenchymal transition, tumor growth, metastasis and chemo-resistance, while its circulating levels correlate negatively with lung function and its genetic variants can produce multisystem developmental disorders.
