  • LRRK2 (leucine-rich repeat kinase 2) is a large, multidomain protein belonging to the ROCO family, featuring a Ras-of-complex (ROC) GTPase domain followed by a C-terminal-of-ROC (COR) domain and a serine/threonine kinase domain, along with armadillo, ankyrin, leucine-rich repeat and WD40 repeat regions that collectively mediate protein–protein interactions, membrane association and autoregulation, and whose pathogenic mutations—most notably G2019S in the kinase domain—are the commonest known genetic cause of both familial and sporadic Parkinson’s disease by promoting hyper-kinase activity, mitochondrial fragmentation, autophagic-lysosomal dysfunction and neuronal toxicity.
