PERK (protein kinase RNA-like endoplasmic reticulum kinase) is an ER-resident transmembrane serine/threonine kinase that, under ER stress caused by misfolded protein accumulation, oligomerizes and auto-phosphorylates to phosphorylate eIF2α, transiently attenuating global cap-dependent translation while selectively up-regulating ATF4 and other stress-response mRNAs, thereby activating a transcriptional program that enhances folding capacity, antioxidant responses, and autophagy, and if stress is unresolved triggers apoptosis, making PERK a central regulator of the unfolded protein response, cancer cell survival, metabolic homeostasis, and neurodegenerative diseases.
