Caspase-2 is an evolutionarily conserved initiator caspase that localizes to the nucleus, cytosol, and Golgi apparatus, where it exists as an inactive 48 kDa pro-enzyme that is activated by dimerization and subsequent autocatalytic processing in response to genotoxic stress, metabolic dysfunction, or mitotic catastrophe, thereby cleaving a limited set of substrates—including MDM2, Bid, and caspase-3—to trigger either a delayed mitochondrial-apoptotic pathway that is independent of p53 or a direct caspase-3 cascade, while also unexpectedly promoting cell survival and genomic stability in some contexts through non-apoptotic functions such as limiting aneuploidy and enhancing DNA repair, illustrating its dual role as both a tumor suppressor and a context-dependent oncogenic regulator.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

ICC shows positive staining in Jurkat cells. Anti-Caspase-2 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
