PPARA (peroxisome proliferator-activated receptor alpha) is a ligand-activated transcription factor of the nuclear hormone receptor family that acts as a master regulator of lipid metabolism, primarily in liver, heart, skeletal muscle and kidney; upon binding fatty acids, eicosanoids or hypolipidemic fibrates it heterodimerizes with RXR, translocates to the nucleus and binds PPREs to activate genes driving peroxisomal and mitochondrial β-oxidation, ketogenesis, gluconeogenesis and lipoprotein remodeling while also exerting anti-inflammatory and anti-ferroptotic effects through cross-talk with GPX4 and other pathways.

ChIP
Chromatin immunoprecipitation (ChIP) was performed on HepG2 cells cross-linked with 1% formaldehyde for 10 min, then chromatin was fragmented by sonication. Parallel reactions used PPARA Recombinant Rabbit mAb (S-3103-24) and Rabbit mAb IgG Isotype Control (SDT-R173) at 1:50 for immunoprecipitation.
Post -immunoprecipitation, both samples were washed, eluted, and cross-links reversed. Purified DNA was analyzed by qPCR.
qPCR showed the enrichment of ERFL, NFZX and SAT-α in PPARA Recombinant
Rabbit mAb (S-3103-24)-immunoprecipitated sample.
