CD94, encoded by KLRD1, is a type II transmembrane C-type lectin glycoprotein expressed predominantly on natural killer (NK) cells and subsets of CD8⁺ T cells, where it covalently assembles with NKG2A, C, E or F subunits to form disulfide-linked heterodimers that recognize the non-classical MHC-I molecule HLA-E (or Qa-1 in mice) and deliver either inhibitory or activating signals that finely tune cytotoxicity, cytokine secretion and survival of effector lymphocytes, thereby playing a central role in innate and adaptive immunity, tumor immune evasion, viral defense and pregnancy tolerance.
流式分析
Flow cytometric analysis of Human PBMC (human peripheral blood mononuclear cells) labelling CD94 antibody at 1/50 dilution (1 μg) / (Right panel) compared with a Rabbit IgG Isotype Control / (Left panel). Goat Anti-Rabbit IgG DyLight 488 was used as the secondary antibody. Then cells were stained with CD56 - Allophycocyanin Antibody separately.
免疫细胞化学
ICC shows positive staining in NK-92 cells (top panel) and negative staining in Jurkat cells (below panel). Anti-CD94 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
