Rictor (rapamycin-insensitive companion of mTOR) is a 1,709-amino-acid, 190 kDa cytosolic scaffold protein that is an obligate core subunit of mTORC2, where it stabilizes the complex and recruits substrates such as AKT, PKCα and SGK1 for phosphorylation by mTOR on key hydrophobic motifs, thereby controlling cell survival, metabolism, cytoskeletal organization, embryonic stem-cell differentiation and lymphatic valve formation, while its frequent overexpression or mutation in cancers links it to tumor growth, invasion and drug resistance.
