PBEF/Visfatin/NAMPT is a multifunctional 52-kDa protein that acts both intracellularly as the rate-limiting enzyme nicotinamide phosphoribosyltransferase (EC 2.4.2.12) in the NAD salvage pathway—catalyzing conversion of nicotinamide to nicotinamide mononucleotide (NMN)—and extracellularly as an adipokine/cytokine circulating in blood; it enhances B-lymphocyte precursor maturation, inhibits neutrophil apoptosis, binds and activates the insulin receptor to exert insulin-mimetic glucose-lowering effects, promotes vascular smooth-muscle cell maturation, modulates endothelial function, and its serum levels correlate with obesity and metabolic syndrome, while intracellularly it maintains the NAD⁺ pool essential for processes such as SIRT1 activity, DNA repair and β-cell glucose-stimulated insulin secretion.

ICC shows positive staining in HeLa cells. Anti-PBEF/Visfatin/NAMPT antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
