  • Phospho-Rb (Ser780) refers to the specific modification form of the retinoblastoma protein (Rb) phosphorylated at its serine 780 residue. The Rb protein is a key regulator of the cell cycle G1/S checkpoint. In its unphosphorylated state, it halts cell cycle progression by binding to and inhibiting E2F transcription factors. When cells receive proliferative signals, the cyclin-dependent kinase 4/6-cyclin D complex specifically catalyzes the phosphorylation of the Rb protein at the Ser780 site. This event is an early and critical step in the inactivation process of the Rb protein. Phosphorylation at Ser780 directly disrupts the interaction between Rb and E2F, thereby relieving the inhibition. The released E2F then activates the transcription of a suite of genes necessary for S-phase entry, driving the cell cycle forward. Consequently, detecting the level of Phospho-Rb (Ser780) is commonly used to assess CDK4/6 activity and the state of cell proliferation, holding significant importance in cancer research and the development of targeted therapies, such as CDK4/6 inhibitors.

检测磷酸化蛋白时,推荐有什么封闭体系?

我们推荐客户使用TPST+5%脱脂奶粉来稀释一抗,进行封闭。 虽然BSA被推荐为WB检测磷酸化蛋白的常用封闭剂,但是脱脂奶粉获取更加方便,覆盖更广泛的非特异性结合位点,在一抗性能优越的前提下,使用脱脂奶粉封闭性价比更高
