GLI1 (Glioma-associated oncogene homolog 1) is an 110 kDa zinc-finger transcription factor of the GLI family that acts as the primary activator of the Hedgehog (Hh) signaling pathway: in response to Hh ligands, smoothened-mediated inhibition of the Patched receptor is relieved, allowing GLI1 to accumulate in the nucleus where it binds GACCACCCA-like motifs to drive expression of target genes that regulate cell proliferation, stem-cell maintenance, differentiation, and tissue patterning during embryogenesis; in adults, tight control of GLI1 is essential because its dysregulation—through activating mutations, ligand overproduction, or loss of suppressors such as PTCH1 or SUFU—promotes oncogenic transcriptional programs implicated in basal-cell carcinoma, medulloblastoma, pancreatic, breast, and other cancers, making GLI1 both a biomarker of pathway activity and a therapeutic target for small-molecule inhibitors like vismodegib and arsenic trioxide.
