GADD34, also known as PPP1R15A, is a stress-inducible regulatory subunit of protein phosphatase 1 (PP1) that plays a crucial role in cellular stress responses by forming a complex with PP1 to dephosphorylate eIF2α, thereby promoting translational recovery during the integrated stress response (ISR), and is involved in various cellular processes including apoptosis, cell cycle arrest, and DNA damage response, with its expression being upregulated by a variety of stress signals such as DNA damage, endoplasmic reticulum (ER) stress, and oxidative stress, and has been implicated in several pathological conditions including cancer, neurodegenerative diseases, and metabolic disorders.
