  • Histone H3 tri-methylated lysine 9 (H3K9me3) is a crucial epigenetic mark formed by the trimethylation of the ninth lysine residue on the core histone H3. It is widely recognized as a classic hallmark of constitutive heterochromatin. Its primary function is to recruit effector proteins like Heterochromatin Protein 1 (HP1), which subsequently promotes the compaction of chromatin into a dense, transcriptionally silent higher-order structure. This modification plays a key role in maintaining genomic stability by effectively repressing the aberrant expression of repetitive genomic elements, such as transposons and telomeric regions, as well as foreign genes. Furthermore, H3K9me3 is essential for the transcriptional regulation of specific genes, X-chromosome inactivation, and cell fate determination. It is important to note that aberrant distribution of H3K9me3 is closely associated with various diseases, including cancer development, neurodegenerative disorders, and the aging process.

  • ChIP

    • Chromatin immunoprecipitation (ChIP) was performed on HeLa cells cross - linked
      with 1% formaldehyde for 10 min, then chromatin was fragmented by sonication.
      Parallel reactions used Histone H3 (tri methyl K9) Recombinant Rabbit mAb (S-3826) and Rabbit mAb IgG Isotype Control (SDT-R173) at 1:50 for immunoprecipitation.
      Post -immunoprecipitation, both samples were washed, eluted, and cross - links reversed. Purified DNA was analyzed by qPCR.
      qPCR (%input: immunoprecipitated DNA/input DNA)
      showed the enrichment of RPL30, GAPDH, MYOD1,
      AFM, SAT-α and SAT-2 in Histone H3 (tri methyl K9) Recombinant
      Rabbit mAb (S-3826)-immunoprecipitated sample.
