Calnexin is an integral endoplasmic reticulum (ER) type I transmembrane lectin chaperone that transiently binds newly synthesized monoglucosylated N-glycoproteins through its luminal carbohydrate-binding domain, coordinates sequential folding events with the oxidoreductase ERp57 to form disulfide bonds, retains incompletely folded or misassembled polypeptides in the ER via a cytosolic phosphorylation-regulated retrieval motif that interacts with the Sec61 translocon, and thereby serves as a central quality-control checkpoint that prevents aggregation and premature trafficking, while also participating in ER calcium homeostasis and cellular stress responses such as the unfolded protein response (UPR).

ICC shows positive staining in HeLa cells. Anti- Calnexin antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
