背景介绍

Tim23 is an essential subunit of the TIM23 translocase in the mitochondrial inner membrane, where it serves as both a structural scaffold and a voltage-gated component that recognizes presequences of incoming precursor proteins in the intermembrane space, triggers channel opening in response to membrane potential, and—together with Tim17, Tim44, mtHsp70 and other motor proteins—forms a highly conserved multiprotein machine that imports ~1,000 matrix-destined and inner-membrane proteins from the cytosol into mitochondria, thereby undergirding organellar biogenesis and cellular energy metabolism.

- ICC shows positive staining in SK-BR-3 cells. Anti- Tim23 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
