背景介绍
  • PLK1 (Polo-like kinase 1) is a 603-residue serine/threonine kinase that peaks in late G2/M where it docks to phosphorylated Bub1 and the PBD-binding p-T78 motif on its own T-loop to auto-activate, then successively phosphorylates Cdc25C, Cyclin B1, Wee1, APC/C-Cdc20, and myriad kinetochore proteins to drive centrosome maturation, bipolar spindle assembly, chromosome alignment, the spindle-assembly and decatenation checkpoints, and timely mitotic exit, while its deregulated overexpression in cancers creates chromosomal instability and chemoresistance, making it a high-priority drug target whose ATP-competitive inhibitors (BI 2536, volasertib) are already in oncology trials.

    • Flow cytometric analysis of 4% PFA fixed 90% methanol permeabilized HeLa (human cervix adenocarcinoma epithelial cell) labelling PLK1 antibody at 1/50 dilution (1 μg)/ (red) compared with a Rabbit monoclonal IgG (black) isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (blue). Goat Anti - Rabbit IgG Alexa Fluor® 488 was used as the secondary antibody.

    • ICC shows positive staining in HeLa cells. Anti-PLK1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
