背景介绍

Peripherin is a 57 kDa type III intermediate filament (IF) protein selectively expressed in the peripheral and enteric nervous systems, where it co-assembles with other IF subunits to form a dynamic scaffold that maintains neuronal shape, supports axonal outgrowth during development and regeneration, and provides resilience against mechanical stress; its expression is markedly up-regulated after axonal injury, making it a widely used marker of neuroplasticity, and pathogenic mutations or deranged aggregation of peripherin has been implicated in motor-neuron degenerative diseases such as amyotrophic lateral sclerosis.

- ICC shows positive staining in Neuro-2a cells. Anti- Peripherin antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
