PKC delta (Protein Kinase C delta) is a serine/threonine protein kinase belonging to the Protein Kinase C family, and its activation is highly dependent on calcium ions and diacylglycerol (DAG) signaling. Unlike other PKC isoforms, PKC delta plays a dual role in cells, promoting both cell survival and apoptosis, with its specific function depending on cell type, subcellular localization, and activation context. It regulates critical cellular processes such as proliferation, differentiation, migration, and DNA damage response by participating in multiple signaling pathways (e.g., NF-κB and MAPK pathways). PKC delta is particularly important in immune regulation, cancer development, and neurological functions. Studies have shown that its abnormal expression or activation is closely associated with tumor progression, autoimmune diseases, and cardiovascular pathologies, making it a potential therapeutic target. The activity of PKC delta is finely regulated by phosphorylation, proteolytic cleavage, and changes in subcellular localization. For example, during apoptosis, it is cleaved by caspases to generate a constitutively active catalytic fragment that amplifies cell death signals.

ICC shows positive staining in HeLa cells. Anti- PKC delta antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
